PROBLEM TO BE SOLVED: To readily, quickly and correctly determine BOD of industrial waste water and other waste water to be treated by determining a luminous amount by assimilation of an organic substance of a fungus having a luminous gene group containing a luciferase gene and a substrate synthesized gene. SOLUTION: A microorganism suspension wherein a fungus (e.g.; Escherichia coli) to which a luminous gene group containing a luciferase gene such as lux A or lux B and a substrate synthesized gene such as lux C-lux E of a luminous substrate such as luciferin are introduced is suspended in a buffer solution is set in a lumi-counter by filling in a glass microtube, a sample of various waster waters is added to the suspension, a luminous amount of the fungus by assimilation of an organic substance is determined by a digital voltmeter as a number of electrons emitted by a photoelectric effect of a photomulticounter in the lumi-counter, the resultant data are transmitted to a personal computer and a data calculation is operated to readily, quickly and correctly determine BOD of industrial waste water and other waste water to be treated. |
