<p>The present invention relates to an anti-inflammatory agent containing, as an active ingredient, (E)-1-(2-(2-nitro vinyl)phenyl)pyrrolidine (NVPP) as an inhibitor of TRIF-dependent signaling pathway induced by toll-like receptors (TLRs). The TLRs perform an important role in the recognition of many photogenic molecule patterns, and induce a congenital immune response. The recognition of bacterial factors by TLRs suggests MyD88 and (TRIF)-dependent downstream signaling pathways. As described above, the present inventors synthesized (E)-1-(2-(2-nitro vinyl)phenyl)pyrrolidine (NVPP) containing nitrovinyl-phenyl and pyrrolidine. In order to evaluate the potential of NVPP as a therapeutic agent, the signaling induction effect of TLRs through TRIF-dependent pathway induced by lipopolysaccharide (LPS), polyriboinosinic polyribocytidilic acid (Poly[I:C]) was monitored. NVPP inhibits activation of an interferon induced gene, such as interferon induced protein-10, and inhibits phosphorylation of NF-kB, interferon regulation factor, and interferon regulation factor 3, induced by LPS or Poly[I:C]. This result suggests that NVPP regulates the TRIF-dependent signaling pathway of TLRs, and thus can be developed as an effective potential therapeutic agent for chronic inflammatory disease.</p> |
