PROBLEM TO BE SOLVED: To provide a biofuel cell capable of drastically improving a current density or an output density by optimizing concentration of an electronic mediator to be immobilized together with an enzyme to a cathode or an anode, and to provide a manufacturing method thereof.

SOLUTION: The biofuel cell has a structure in which a positive electrode 2 and a negative electrode 1 are opposed to each other via an electrolyte layer 3 provided therebetween, at least either the positive electrode 2 or the negative electrode 1 made of an electrode on which at least one kind of enzymes and at least one kind of electron mediators are immobilized. A concentration of the electron mediator immobilized on the electrode is at least 10 times a Michaelis constant Kmof the enzyme of the electron mediator, which is determined by measurement in solution.
