PROBLEM TO BE SOLVED: To provide a measuring reagent of a biological substance, which suppresses the deterioration in measurement sensitivity with time even if being a highly alkaline reagent. SOLUTION: A measuring reagent of a biological substance includes a highly alkaline buffer reagent containing a volatile water-soluble solvent. The volatile water-soluble solvent is at least one selected from a group which comprises ethanol, methanol, 1-propanol, 2-propanol, acetone, acetonitrile, and tetrahydrofuran, and can be used for a Ca measurement by an o-CPC method or an alkaline phosphatase measurement.
