DNA having a Y-shaped structure in the present invention binds to TLR9 to effectively induce an acquired immune reaction by stimulating congenital immune cells such as increasing activities of IFN-βand COX2 promotor, increasing secretion of IL-12 and TNF-αof dendritic cells, and increasing expression of CD80 and CD86 which are auxiliary stimulation polymers on the surface of dendritic cells. The DNA having a Y-shaped structure according to the present invention can be used as a composition for immune boosting or a pharmaceutical composition for preventing or treating immunization diseases, and also can be used as a composition for increasing anticancer effect which can increase effects of an anticancer agent by being administered with an anticancer agent.
