Embodiments of compounds for selectively detecting a thiol are disclosed. In some embodiments, the compounds are bridged viologens, and the compounds are capable of reacting with homocysteine and/or glutathione in a buffered solution to produce a change in the solution's absorbance spectrum and/or emission spectrum. Also disclosed are embodiments of methods and kits for detecting homocysteine and/or glutathione with the disclosed bridged viologens.

1. A compound having:
(a) Formula Iwhere R is a lower alkyl nitrile, alkynyl, 2-oxo-2-(2-oxo-2H-chromen-3-yl)ethyl, 2,2-dihydroxyethyl, carboxymethyl, or a fluorophore; or
(b) Formula IIwhere R is a lower alkyl nitrile, alkenyl, alkynyl, 2-oxo-2-(2-oxo-2H-chromen-3-yl)ethyl, 2,2-dihydroxyethyl, carboxymethyl, or a fluorophore; or
(c) Formula IIIwhere R is a lower alkyl nitrile, alkenyl, alkynyl, 2-oxo-2-(2-oxo-2H-chromen-3-yl)ethyl, 2,2-dihydroxyethyl, carboxymethyl, or a fluorophore,
wherein the fluorophore has the structure: where X is oxygen, sulfur, CH2 or NH; R1, R2, R3, R4, and R6 independently are —H, —OH, lower alkyl, carboxyalkyl, amino, substituted amino, alkoxy, or halogen, or R2 and R3 together may form a substituted or unsubstituted cycloalkyl or aryl; R5 is oxygen, imino, substituted alkyl imino, or substituted or unsubstituted cycloalkyl imino; R7 is —H, alkyl, acyl, carboxyl, nitro, amino, substituted amino; and R8 is absent or alkyl.
