METHOD FOR SCREENING YEAST STRAINS WITH ENHANCED RECOMBINANT PROTEIN SECRETION USING YEAST VARIANTS HAVING CELL-WALL DEFECT

The present invention relates to a method for screening yeast mutant secreting induced target protein, a method for increasing ability of secreting the target protein, a method for manufacturing the target protein, and a method for manufacturing yeast strain with enhanced Gas1 or fusion protein secretion ability. The yeast strain of the present invention expressing fusion protein having a target protein, which is manufactured by using GAS1-lacking strain in which the growth is inhibited under a cell wall stressed condition, bonded to N-terminal of Gas 1 protein is identified to have growth ability increased by forming cell wall according to secretion of the fusion protein to effectively identify yeast strain with increased secretion ability. The yeast strain can be used to select a strain with increased secretion from a series of mutant library manufactured by random mutation for mass production of recombinant protein.
