In the present invention, the function of Rv3168 protein, which was not known, is firstly explained with resistance to kanamycin. Provided are a method and a kit for detecting mycobacterium tuberculosis with resistance to kanamycin wherein the method and kit are allowed to detect whether mycobacterium tuberculosis has resistance to kanamycin as an anti-tuberculous agent. Therefore, the method and the kit of the present invention can be used for diagnosing tuberculosis and for selecting drugs when treating tuberculosis.
