The present invention relates to a non-viral drug delivery carrier composition containing a nucleotide and a non-viral drug delivery carrier, wherein the composition has proton buffering activity or hemolysis activity thereby inducing endosomal escape of a drug to be delivered through endosomolysis. The non-viral drug delivery carrier composition according to the present invention is related to an effective drug delivery carrier capable of enabling the escape of the drug that is delivered from an endosome into other organelles in a cell by means of the proton buffering activity or the hemolysis activity of the nucleotide.
