PROBLEM TO BE SOLVED: To provide an integrase-defective lentiviral donor polynucleotide for expression of a heterogenous polynucleotide.SOLUTION: An integrase-defective lentiviral donor polynucleotide is selected from the group consisting of: i) a donor polynucleotide comprising a target gene located between a first and second homologous sequences and a first selection marker allocated outside the first and second homologous sequences; ii) a donor polynucleotide comprising a target gene and a first selection marker allocated between the first and second homologous sequences and a second selection marker allocated outside the first and second homologous sequences, iii) a donor polynucleotide comprising a target gene and a first selection marker allocated outside the first and second homologous sequences, and iv) a donor polynucleotide comprising a target gene allocated between the first and second homologous sequences and a selection marker, where the first and second homologous sequences are identical to endogenous chromatin sequences of the mammalian cell. |
