The present invention relates to a cell stimulation device and a stimulation method thereof using a three dimensional rotating device and ultrasonic waves and, more specifically, the cell stimulation device comprising a rack; a supporting member whose one end is connected to the upper surface of the rack; an external frame as a square-shaped frame, in which four members are connected, which rotates with respect to one axis direction in two separated supporting members; and an inner frame as a square-shaped, in which four members are connected, which is installed to be rotated inside with respect to an axis direction crossed with the rotation direction of the external frame.
