The present invention relates to organophosphate-inhibited acetylcholinesterase (AChE) reactivators with central nervous system (CNS) bioavailability and their associated methods of preparation including their use for treatment of exposure to AChE inhibitors such as an organophosphate (OP) insecticides and nerve agents.

An acetylcholinesterase reactivator comprising the formula: wherein R1 is a ketone functionality (C(O)R) or hydrogen and R may be a H or an alkyl group containing up to five carbon atoms; R2 is a hydrogen or an alkyl group containing up to five carbon atoms; R3 may be H or an electron withdrawing group or electron donating group; Y may be a carbon or nitrogen; and n has a value of 0-4.
