A modified human tumor necrosis factor receptor-I (TNFRI) polypeptide or fragment thereof comprising an amino acid sequence selected from the group consisting of: i) the amino acid sequence of natural TNFRI as set forth in SEQ ID NO: 1; ii) the amino acid sequence composed of amino acid residues 41 to 211 of the amino acid sequence of the natural TNFRI set forth in SEQ ID NO: 1 (TNFRI171); iii) the amino acid sequence composed of amino acid residues 41 to 166 of the amino acid sequence of the natural TNFRI set forth in SEQ ID NO: 1 (TNFRI126); and iv) the amino acid sequence composed of amino acid residues 41 to 145 of the amino acid sequence of the natural TNFRI set forth in SEQ ID NO: 1 (TNFRI105); wherein the amino acid sequence is modified at least at positions 92, 95, 97 and 98.
