Provided in the present invention are a gene tsh-Ser for preventing colibacillosis, a protein encoded thereby and the preparation method thereof. The method comprises: using 5'-atgaacagaatttatt-3' as an upstream primer zxl, 5'-gaatgaataacgaa-3' as a downstream primer zx2, and the plasmid DNA of a O78 serotype Escherichia coli as a template, and using a PCR amplification method to obtain the gene tsh-Ser for preventing colibacillosis, and then using the gene to obtain the protein encoded thereby.
