PROBLEM TO BE SOLVED: To provide an industrial enzymatic saccharification treatment method of lignocellulosic biomass, which enables recovery of used enzyme from a reaction liquid at a high recovery rate and cyclic use thereof over a long period of time. SOLUTION: In the enzymatic saccharification treatment method of a lignocellulosic raw material, calcium carbonate particles are added to a raw suspension that contains the lignocellulosic raw material, which is pretreated to make it a raw material suitable for an enzymatic saccharification reaction, to perform the enzymatic saccharification reaction, the enzyme-containing liquid is separated and recovered from the treated suspension, and the recovered enzyme-containing liquid is circulated to an enzymatic saccharification treatment process. Also, in the enzymatic saccharification treatment method, fermentation microorganisms, whose fermentation substrate is generated saccharides, are added to the raw suspension to perform the enzymatic saccharification reaction and the fermentation reaction of the saccharides at the same time.
