In the present invention, provided are 3-(4-amino-1-oxo-1,3 dihydro-isoindol-2-yl)-piperidine-2,6-dione, a novel polymorph J,K, and L of lenalidomide which is a pharmaceutical composition for reducing the concentration of a tumor necrosis factor a, and a manufacturing method thereof. The novel polymorphs of the present invention have greater physical and chemical properties such as thermal stability and water solubility than conventional polymorphs.
