In the present invention, there is described a method of storing DNA from histopathological material characterized in that the histopathological material is applied to a microscope slide intended for analysis of individual cells. Subsequently, a lysis buffer L, prepared by mixing buffer T containing 30 mM of Tris-HCl and having pH = 8, and a solution K, containing proteinase K in concentration of 20 mg/ml in the buffer and composition of 20 mM Tris-HCl, 1mM CaCli2, 50 percent glycerol, and having pH = 7.3, in a ratio 2T:1K to 5T:1K are added to the applied histopathological material such, as to form a drop onto the surface of the histopathological material intended for isolation of the DNA. The so formed drop is then completely covered by mineral oil and subjected to incubation for a period in the range of 1 to 12 hours at a temperature ranging from 50 to 60 degC followed up with another incubation for a period of 5 to 15 minutes at a temperature ranging from 75 to 95 degC. Subsequently, the half amount of the added mineral oil is removed by pipetting and the remaining sample is dried in the slide at a temperature ranging from 40 to 70 degC. The invention also describes a preferred method of sampling the DNA being stored. |
