A method for detecting a mutation, said method comprising steps for: contacting a DNA sample which is likely to have a mutation with a first query probe having a first discrimination sequence, by which the mutation can be discriminated, and a second query probe having a second discrimination sequence, by which a region adjacent to the mutation can be discriminated, and conducting a DNA ligase reaction to give a ligation product that has the first discrimination sequence, the second discrimination sequence, a hybridization sequence which is specifically hybridizable with a sequence for detection of a probe depending on the type of the aforesaid mutation, and a labeling sequence for imparting a label; contacting the ligation product with a first primer having a sequence complementary to the hybridization sequence and a second primer having the labeling sequence, and conducting an amplification reaction using DNA polymerase to give an amplification product that has the labeling sequence and the hybridization sequence; and contacting the amplification product with the probe on a solid phase carrier to give a hybridization product, wherein the total concentration of the second primer is 0.5 (inclusive) to 12.5 (exclusive) times as high as the total concentration of the first primer. |
