The subject matter disclosed herein pertains to a method for screening drugs using second-harmonic generation microscopy. The tissue is scanned with a pulsed laser light which has an excitation wavelength. At least some of microtubules within the tissue produce generated light with a second-harmonic wavelength that is half the excitation wavelength. A microtubule pattern within the tissue is determined based on an analysis of the generated second-harmonic wavelength. |

1. A method for screening drugs in a retinal tissue using label-free second-harmonic generation microscopy, the retinal tissue having a retinal nerve fiber layer comprising microtubules, the method comprising the steps of:
exposing the retinal tissue to a drug; emitting a pulsed laser light having an excitation wavelength and a predetermined first polarization or a predetermined first spatial mode; scanning the retinal tissue with the pulsed laser light, at least some of the microtubules producing generated light with a second-harmonic wavelength that is half the excitation wavelength; measuring a generated polarization or a generated spatial mode of the generated light; determining the difference between (a) the first polarization and the generated polarization or (b) the first spatial mode and the generated spatial mode; and determining whether or not the drug impacts the retinal tissue based on the difference. |
