The present invention relates to the field of protein engineering, and relates to a kind of mutated proteins A with a high alkali resistance feature and an application thereof. The proteins A of the present invention can be firmly combined to other regions except a complementarity determining region, of immunoglobulin molecules, and can be used as a ligand coupled to a solid phase carrier to separate immunoglobulins. The kind of mutated proteins A can keep chemical stability in a strong alkali environment with PHs 13 to14, and can be used as a chromatographic ligand to sustain rigid conditions of cleaning in place. The present invention also relates to a process of immunoglobulin separation and purification by using the protein A ligand, and a process of alkali regeneration.
