PROBLEM TO BE SOLVED: To provide a DNA affinity substance-containing composition, a DNA affinity substance-containing composition manufacturing method, a DNA affinity substance purification method, and a DNA affinity substance purification kit which can be used with a high degree of reliability in a DNA amplification method and the like using a trace amount of DNA as a template.SOLUTION: A DNA affinity substance-containing composition contains 0 ag or more to less than 30 ag of a DNA amplifiable in a strand-displacement-type DNA amplification method, per 100 ng of a DNA affinity substance in the composition. A DNA affinity substance purification method includes a combination of steps of: removing a DNA contained in a mixture containing the DNA affinity substance from the mixture by causing a DNA absorbent to absorb the DNA; and inactivating the DNA amplification capacity contained in the mixture by using a DNA inactivation agent.
