The present invention relates to a roll cleaning system for preventing contamination of the rear side in roll-to-roll printing and, more specifically, to a roll cleaning system for preventing contamination of the rear side in roll-to-roll printing to clean ink transferred to a nip roll for generating contamination in the rear side of a material when roll-to-roll continuous printing is performed. According to the present invention, the roll cleaning system for preventing contamination of the rear side in roll-to-roll printing comprises a printing roll having a fixed printing pattern on an outer circumferential surface; a nip roll adjacent to the printing roll; and a cleaning part for providing a cleaning material to remove ink transferred to the nip roll due to punching of a transfer material by interposing the cleaning material between the printing roll and the nip roll.
