Formulations and methods of manufacture are provided for granisetron dry powder compositions suitable for intranasal administration. Also provided are methods of use for preventing or controlling emesis and other diseases and disorders and devices, compositions, and methods for nasal delivery of therapeutic formulations. Devices for delivery of dry powder formulations are also provided. Devices can be single-use devices.

1. A device comprising:
a) a nozzle having an upstream end and a downstream end adapted to allow positioning of at least a portion of the nozzle into a nostril of a subject, the nozzle having a reservoir, wherein the reservoir comprises a single dose of a powdered therapeutic formulation that is disposed within the reservoir of the nozzle; b) a valve assembly having an upstream end and a downstream end, wherein the downstream end is coupled to the upstream end of the nozzle; and c) an air source operably linked to the upstream end of the valve assembly, wherein when the device is not in use, the reservoir is in communication with the valve assembly through an opening or a duct, and the valve assembly comprises a poppet adapted to block the movement of the powdered therapeutic formulation into the valve assembly.
