PROBLEM TO BE SOLVED: To provide a free base of 6-acetyl-8-cyclopentyl-5-methyl-2-(5-piperazine-1-yl-pyridine-2-ylamino)-8H-pyrido[2,3-d]pyrimidine-7-one demonstrating improved physicochemical and manufacturing possibility properties and having larger primary particle diameter.SOLUTION: The invention relates to a crystalline free base of 6-acetyl-8-cyclopentyl-5-methyl-2-(5-piperazine-1-yl-pyridine-2-ylamino)-8H-pyrido[2,3-d]pyrimidine-7-one having improved properties, a pharmaceutical composition and a formulation containing the free base, and methods of manufacturing such compounds, compositions and formulations, and a method of using them in treatments of cell proliferative diseases such as cancer. |
