PROBLEM TO BE SOLVED: To provide a cell harvesting method from the packed-bed system, which can expand easily to a practical production scale.SOLUTION: A large scale cell harvesting method comprises steps of: providing a pack-bed culture device configured with a culture chamber having a cell growing space defined within the culture chamber; performing a cell detaching step at least comprising shaking or spinning the culture chamber; and harvesting the plurality of cells, wherein the cell growing space is loosely packed with a plurality of porous matrixes, the plurality of cells grow on the porous matrixes, and a void volume that is unoccupied space defined by the porous matrixes in the cell growing space is large enough for the porous matrixes to collide with each other or with the culture chamber and to generate momentum by which the cells are detached from the porous matrixes.
