Producing 3-hydroxyisobutyric acid, comprises: (a) contacting isobutyric acid with a combination of isobutyrate kinase and phosphotransisobutyrylase and/or isobutyryl-coenzyme A-synthetase/ligase and/or isobutyrate-coenzyme A-transferase; (b) contacting the product obtained from the step (a) with isobutyryl-coenzyme A-dehydrogenase; (c) contacting the product obtained from the step (b) with methacrylyl-coenzyme A-hydratase; and (d) hydrolyzing the product obtained from the step (c) with the formation of 3-hydroxyisobutyric acid. Producing 3-hydroxyisobutyric acid, comprises: (a) contacting isobutyric acid with a combination of isobutyrate kinase and phosphotransisobutyrylase and/or isobutyryl-coenzyme A-synthetase/ligase and/or isobutyrate-coenzyme A-transferase; (b) contacting the product obtained from the step (a) with isobutyryl-coenzyme A-dehydrogenase; (c) contacting the product obtained from the step (b) with methacrylyl-coenzyme A-hydratase; and (d) hydrolyzing the product obtained from the step (c) with the formation of 3-hydroxyisobutyric acid, where at least one of the steps of (a), (b) and (c) is preformed using enzymes in the form of a cell, the enzymes comprise the isobutyrate kinase, phosphotransisobutyrylase, isobutyryl-coenzyme A-synthetase/ligase, isobutyrate-coenzyme A-transferase or an activity of 3-hydroxyisobutyric acid dehydrogenase or a variant of the 3-hydroxyisobutyric acid dehydrogenase, and the activity is reduced with respect to the wild type of the cell. Independent claims are included for: (1) a cell, comprising the enzyme consisting of isobutyryl-coenzyme A-synthetase/ligase, isobutyrate-coenzyme A-transferase, isobutyrate-kinase, phosphotransisobutyrylase, isobutyryl-coenzyme A-dehydrogenase, methacrylyl-coenzyme A-hydratase or 3-hydroxisobutyryl-coenzyme A-hydrolase and having the activity of 3-hydroxyisobutyric acid dehydrogenase or a variant of the 3-hydroxyisobutyric acid dehydrogenase, where the activity is reduced with respect to the wild type of the cell; and (2) a reaction mixture comprising the cell, and isobutane or isobutyric acid. |
