The present invention relates to a method for measuring the activity of protein kinase. The method comprises the steps of: a) coupling N-[γ-maleimidobutyryloxy]sulfosuccinimide ester (GMBS) on a substrate; b) producing a kit for measuring the activity of protein kinase by coupling a substrate reacting with protein kinase with the substrate coupled with the GMBS; c) introducing a mixture of a sample to be analyzed and a buffer including triton X-100 to the kit; and d) measuring the activity of protein kinase by probing the phoshorylation of the substrate by means of the protein kinase include in the sample.
