PROBLEM TO BE SOLVED: To provide a new endo-&bgr;-N-acetylglucosaminidase (Endo-Om) and its gene.SOLUTION: Provided is a new endo-&bgr;-N-acetylglucosaminidase (Endo-Om), by using a strain transformed with the cloned gene of endo-&bgr;-N-acetylglucosaminidase (Endo-Om) derived from methanol-assimilating Ogataea minuta IFO10746. The Endo-Om has a specific activity 13-fold higher and Vmax 55-fold higher than the known Endo-M, being useful for analysis of sugar chain structures and modification of sugar chains including complex-type sugar chains of glycoproteins. Further, based on the gene sequence of Endo-Om, other endo-&bgr;-N-acetylglucosaminidases, of which enzyme activities of cleavage and transfer of complex-type sugar chain are similar to Endo-Om, were isolated from Candida parapolymorpha DL-1 ATCC26012, Pichia anomala ATCC36904, and Zygosaccharomyces rouxii ATCC2623, that is, Endo-Cp, Endo-Pa and Endo-Zr, respectively. |
