Disclosed is a process for the preparation of tetraiodothyronine (T4) / thyroxine (T4) and its salts comprising reacting the compound of formula I (diiodothyronine) with an iodinating agent comprising sodium iodide (NaI) and iodine, in the presence of an aliphatic amine (such as ethylamine), to obtain the disodium salt of tetraiodothyronine (formula II) as a precipitate recovered as a crude; a) preparing a hydroalcoholic alkaline solution of the crude compound of formula II, having a pH of at least 11; b) heating at a temperature of 40ºC to 80ºC; c) cooling to a temperature of 5ºC to 20ºC; and d) isolating compound II as a pure solid. The above process may also further comprise resuspending the compound of formula II in an alkaline aqueous solution, adding an organic or inorganic acid (preferably acetic acid) to the aqueous solution, to a pH of 9-11 and isolating the resultant mono-sodium derivative. |
