一种利用基因芯片检测跌打镇痛膏药效的方法,其特征在于依次包括如下步骤:A:取一家兔,然后采用重锤坠落法制备软组织损伤模型:选用重量为1kg的重锤,从家兔后退根部软组织的垂直上方20cm处自由落下,获得软组织损伤模型;步骤B:在A步骤完成的24小时后,测量家兔软组织损伤评分,其中测定痛阀采用的仪器为压痛仪,红肿范围不小于1平方厘米;步骤C:在家兔的损伤部位贴上跌打镇痛膏;步骤D:在家兔的取损伤部位处提取RNA,备用;步骤E:选用DNA芯片,与步骤D提取的RNA进行原位杂交,其中所述DNA芯片具有针对如下基因产生的RNA、cRNA或cDNA进行检测的探针:1)炎症反应相关:chemokine(C‑C motif)ligand11、class II transactivator、interleukin1beta、selectin,platelet,tumor necrosis factor receptor superfamily,member1b;2)创伤应答相关:chemokine(C‑C motif)ligand11、class II transactivator、ypoxia‑inducible factor1,alpha subunit(basic helix‑loop‑helix transcription factor)interleukin1beta、selectin,platelet、tumor necrosis factor receptor superfamily,member1b;3)免疫应答相关:chemokine(C‑C motif)ligand11、class II transactivatorinterleukin1beta、phospholipid scramblase1、tumor necrosis factor receptorsuperfamily,member1b;4)防御应答相关:chemokine(C‑C motif)ligand11、class II transactivator、hypoxia‑inducible factor1、alpha subunit(basic helix‑loop‑helix transcriptionfactor)、interleukin1beta、selectin,platelet、tumor necrosis factor receptorsuperfamily,member1b;5)血管生长相关:Kruppel‑like factor5、hypoxia‑inducible factor1、alphasubunit(basic helix‑loop‑helix transcription factor)、interleukin1beta、selectin,platelet;6)能量相关:ATPase,Ca2+transporting,cardiac muscle,slow twitch2、class IItransactivator、hexokinase3(white cell)、myosin,heavy chain7B,cardiac muscle,beta phosphofructokinase,muscle phosphofructokinase,platelet,runt relatedtranscription factor1、sulfotransferase family,cytosolic1A,phenol‑preferring,member1;步骤F:采用基因芯片扫描仪进行扫描、获取扫描信号和数据,最后将信号和数据利用Quantile算法进行归一化处理。 |
