PROBLEM TO BE SOLVED: To provide screening methods for obtaining enzymes each having one or more activities among an RNA-dependent DNA polymerase activity, a DNA-dependent DNA polymerase activity, and a ribonuclease H activity, the enzymes being improved in expression level and thermal stability.SOLUTION: A screening method includes a step of introducing mutation into a polynucleotide encoding an enzyme which has one or more activities among the three activities, a step of inserting the polynucleotide into which the mutation has been introduced, a step of genetically transforming a host with the expression vector into which the polynucleotide has been inserted, a step of cultivating the obtained transformant to express the enzyme, a step of extracting the enzyme from the culture medium of the transformant, and a step of evaluating the performance of the enzyme by a nucleic acid synthesis. |
