PROBLEM TO BE SOLVED: To provide a method and a composition for polymer-delivery enhancement.SOLUTION: The present invention provides a composition and method for enhancing tight junction-across delivery of a large polymer (i.e., over 10 kDa) such as antigen-binding polypeptide. Such a method and composition is particularly useful for delivery of a therapeutic antigen-binding polypeptide to CNS by intranasal administration in order to treat neurologic disorders. In one aspect, the present invention provides a composition comprising one or more antigen binding polypeptides such as immunobinder (e.g., scFv) and one or more permeation enhancing agents (e.g., Pz peptide or FMOC peptide). In a certain embodiment, the antigen binding polypeptide is covalently bound to a permeation enhancing agent.
