Expression of Tomato yellow leaf curl virus(TYLCV)-C2 gene in Escherichia coli and production of polyclonal antibody against the recombinant TYLCV-C2 protein

PURPOSE: A method for preparing polyclonal antibody for TYLCV-C2-6X His fusion protein is provided to develop a virus diagnosis kit. CONSTITUTION: A method for preparing TYLCV(tomato yellow leaf curl virus)-C2-6X His fusion protein comprises: a step of cloning TYLCV-C2 gene to pET28a vector; a step of transforming into E.coli; a step of selecting transformed E.coli colony and culturing; a step of adding IPTG for expression of the recombinant protein and determining optimal condition; a step of isolating and purifying recombinant C2 protein; and a step of performing Ni-NTA resin chromatography and eluting TYLCV-C2-6X His fusion protein.
