The method involves holding a substrate (5) on a sample holder (10). A stationary docking element (7) is arranged relative to the substrate. A specimen (3) is carried in liquid form such that a component (8) of the sample is interacted with the docking element. A visible optical structure of the substrate is formed by the docking element. The component of the sample is identified by image processing. The component is identified at the docking element to automatically focus of an X-ray laser (2). The sample holder is adjusted to achieve a flowing of the sample on the substrate. An independent claim is also included for a device for preparing a sample for performing invitro diagnosis by an X-ray radiation unit.
