The present invention relates to a water-treatment separation membrane including a porous supporting body, a polyamide layer formed on the porous supporting body, and a protective layer formed on the surface of the polyamide layer and containing a compound denoted by chemical formula (I); and a manufacturing method thereof. In the chemical formula (I), L1 is an amine group, haloalkyl, haloalkenyl, haloalkynyl, a hydroxy group, a ketone group, an aldehyde group, an acyl halide group, a carbonate group, an ester group, an ether group, a imine group, an azid group, a nitrile group, a tiocyanate group, a thione group, a thial group, a phosphate group, an epoxide group, an acrylyl amine group, or a thiol group.
