FIELD: chemistry.SUBSTANCE: invention relates to biotechnology and can be used in cardiology to study formation and rapture of plaque in controlled laboratory conditions, observing the life of various cells in the plaque in given conditions and performing comparisonandIn the method of cultivating atherosclerotic plaque explants, the explant material used is portions of carotid arteries obtained from operations performed for obliterating atherosclerosis of carotid arteries, which are stored in the laboratory for not more than 2 hours after the operation; after extraction, the obtained material is washed in a phosphate-salt buffer, after which the cultivating material is cut into annular blocks with thickness of 2-3 mm perpendicular to the aperture of the vessel; 2 blocks are then fixed in 2% paraformaldehyde and enclosed in paraffin; the remaining blocks are placed in the cultivating medium. Tissue is cultivated for not less than 20-22 days; every three days, tissue samples are fixed in 2% paraformaldehyde and then enclosed in paraffin; at the last day of cultivation, the remaining samples are also fixed in 2% paraformaldehyde, enclosed in paraffin and analysed histologically.EFFECT: invention enables to keep atherosclerotic plaque explants alive during cultivation for further study of atherosclerosis pathogenesis mechanisms.1 ex |
