摘要 PROBLEM TO BE SOLVED: To provide a high throughput method for the identification and detection of molecular markers, in which restriction fragments are generated and suitable adaptors comprising (sample-specific) identifiers are ligated.SOLUTION: The adapter-ligated restriction fragment may be selectively amplified with an adaptor compatible primer carrying selective nucleotide at its 3' terminal. The amplified adapter-ligated restriction fragment is, at least partly, sequenced using a high throughput sequencing method and the sequence part of the restriction fragment together with the sample-specific identifier serves as a molecular marker.
