摘要 PROBLEM TO BE SOLVED: To provide immunoglobulins into which novel antigen-binding sites are introduced, and a method to design and produce the immunoglobulins.SOLUTION: A method for engineering an immunoglobulin comprising at least one modification in a structural loop region of the immunoglobulin and determining the binding of the immunoglobulin to an epitope of an antigen, wherein the unmodified immunoglobulin does not significantly bind to the epitope, comprises: the step of providing a nucleic acid encoding an immunoglobulin comprising at least one structural loop region, the step of modifying at least one nucleotide residue of at least one of the structural loop regions, the step of transferring the modified nucleic acid to an expression system, the step of expressing the modified immunoglobulin, the step of contacting the expressed modified immunoglobulin with an epitope, and the step of determining whether or not the modified immunoglobulin binds to the epitope. There is also provided the modified immunoglobulin.
