Mitochondrial expression vector comprising a gene to be expressed and/or a selection marker, and a mitochondrial promoter region is new. Independent claims are included for: (1) inserting DNA to be expressed in the mitochondria of mammalian cells, comprising constructing a mitochondrial expression vector or a mitochondrial genome, performing reversible induction of mega-mitochondria and transfecting the mega mitochondria by a physical transfection method; and (2) reversible induction of mega mitochondria of mammalian cells by acidification of mammalian cells culture medium by addition of sodium acetate or lactic acid. |
