PROBLEM TO BE SOLVED: To provide novel compositions comprising polymeric moieties covalently attached to therapeutic agents in which the therapeutic agent is attached to the polymeric moiety through a tether, methods for controlled delivery of the therapeutic agents by selecting from a variety of tether groups and targeting ligands the polymers, and methods of treating subjects with therapeutic compositions.SOLUTION: The present invention is related to novel compositions comprising polymeric moieties covalently attached to therapeutic agents in which the therapeutic agent is attached to the polymeric moiety through a tether. The present invention is also related to methods for controlled delivery of the therapeutic agents by selecting from a variety of tether groups and targeting ligands the polymers, and methods of treating subjects with the therapeutic compositions described in this specifications.
