PROBLEM TO BE SOLVED: To solve a problem in conventional custom-made genetic diagnosis of hematopoietic tumor and realize ready-made genetic diagnosis of hematopoietic tumor in general clinical practice.SOLUTION: With respect to IgH gene, a V-D-J conjugate (CDRIII) is made after reconstitution of a basal VDJ region. To detect the conjugate, JH probes (LJH, VLJH) to J region is used in the southern method, and Semi-Nested PCR method in which a primer FR3A to V region and primers LJH and VLJH to J region are used is adopted in PCR method. With respect to TCR gene, a V-D-J conjugate (Vδ1-D-Jδ1) is made after reconstitution of the basal VDJ region. To detect the conjugate, Nested PCR method in which a primer D3 to Vδ1 in V region, a primer D4 to Jδ1 in J region, a primer D1 to an outward region of V region and a primer D6 to an outward region of J region are used is adopted. Thereby, it is provided an automated examination method and an examination kit resulting in realizing of ready-made genetic diagnosis in general clinical practice. |
