PURPOSE: A pharmaceutical composition containing tramadol or pharmaceutically acceptable salt thereof is provided to prevent quick release of active ingredients with minimized side effect. CONSTITUTION: A pharmaceutical composition comprises: a core containing tramadol or pharmaceutically acceptable salt thereof; and a plurality of microspheres containing a polymer coating layer for controlling release. The polymer of the polymer coating layer comprises different first and second microspheres. The average thickness ratio of the coating layer of the first and second microspheres is 1: 1.2-8. The coating layer contains water insoluble polymers.
