Disclosed are a white blood cell classifying and counting agent and a method for white blood cell classifying and counting, and applications of an amphoteric surface active agent. The white blood cell classifying and counting agent comprises at least one cationic surface active agent, at least one amphoteric surface active agent and a pH buffer, wherein the amphoteric surface active agent is an amphoteric surface active agent as shown in the general formula I herein or an amphoteric surface active agent of a bile acid derivative type. The white blood cell classifying and counting agent can be used to specifically treat eosinophil granulocytes and enhance the difference between eosinophil granulocytes and neutrophil granulocytes so as to achieve an improvement in the classification of eosinophil granulocytes, and ensure that the difference in classification effect for each subtype of white blood cell between aged blood and fresh blood falls into an acceptable range, thereby making the white blood cell classifying and counting method accurate and quick.
