The invention relates to experimental medicine and laboratory engineering, in particular to reproduction of endothelium physiological activation mechanism for simulating endothelium-dependent processes, such as inflammation, blood clot organization and thrombolysis in bloodstream. According to the method disclosed an isolated vessel fragment is washed with buffer solution and fixed in the closed circulation system. Thereafter the direct perfusion of inner vessel fragment surface is performed with preserved endothelium monolayer first with biosolution, then with addition of proinflammatory biochemical agents, e.g. hydrogen peroxide or thrombin. The process is controlled by dynamics of interleukin-6 perfusate accumulation. The device disclosed comprises a coupling to which the vessel fragment fixed on connectors is inserted and which is air-tightly fixed at outer ends of the connectors, to provide direct perfusion of inner vessel fragment surface device elements are connected with formation of closed circulation system by means of elastic tubes, and container with perfusate is connected to outlet connector. |
