PROBLEM TO BE SOLVED: To provide an adeno-associated virus (AAV) which exhibits tropism for a desired target cell or tissue and is suitable for delivery of a heterologous gene.SOLUTION: The invention provides a method of generating a recombinant adeno-associated virus (AAV) having an adeno-associated virus (AAV) capsid, comprising culturing a host cell which contains (a) a molecule encoding an AAV capsid protein; (b) a functional rep gene; (c) a minigene composed of AAV inverted terminal repeats (ITRs) and a transgene; and (d) sufficient helper functions to permit packaging of the minigene into the AAV capsid protein.
