The present invention discloses a detection method and a detection kit for DNA fragments, and the use thereof. The method comprises the following steps: design primers based on sites or regions to be detected in DNA fragments, cyclize the DNA fragments to obtain cyclized DNA, amplify the cyclized DNA using the primers by PCR, and detect the products of the amplification. In the present invention, amplification can proceed provided that only one of primers matches the template after the cyclization of DNA fragments.
