[Problem] To achieve both measurement reliability and a reduction in measurement time in a coloration measurement device and method. [Solution] A determination process is performed for the presence/absence of a test substance before a cleaning step for supplying to a specimen fragment a cleaning liquid for cleaning a test region and the region peripheral thereto, when the test substance is detected, the determination process is ended, and when the test substance is not detected, the determination process is performed again after performing cleaning.
