A cell counting device and a manufacturing method thereof are provided. The cell counting device comprises: a conductive substrate comprising a standard area and sensing areas are on both sides thereof; a protrusion standard organic layer on the standard area; probes placed on the sensing organic layer of each sensing area; and a detection electrode arranged on the upper part of the probes of each sensing area.
