PURPOSE: A novel exoglucanase which is useful for bio-ethanol production and a manufacturing method of the bio-ethanol are provided to enhance cellulose decomposition activities. CONSTITUTION: A novel exoglucanase is represented by the sequence number 22(SEQ ID NO:22) or by the amino acid sequence of 24. The exoglucanase is revealed from penicillium calcoaceticus. The polynucleotide which codes the exoglucanase is represented by the base sequence of 25 or by the sequence number 23. The expression vector includes the polynucleotide which codes the exoglucanase. The expression vector additionally includes translational fusion partner(TFP). A manufacturing method of the exoglucanase comprises the following steps: cultivating transformant in which the expression vector is introduced to a host cell; and collecting exoglucanase from the culture material or supernatant. A manufacturing method of bio-ethanol comprises the following steps: obtaining transformant in which the expression vector is introduced to the host cell having ethanol fermentation capacity; cultivating the obtained transformant in a culture medium which contains exoglucanase substrate; and collecting the bio-ethanol from the obtained culture materials or supernatant.
