PROBLEM TO BE SOLVED: To provide a separation purification device and method capable of continuously carrying out a process of chromatography with a plurality of steps, while continuously injecting a sample.SOLUTION: In a separation purification device, repeating constitutional units are repeatedly connected. Each of the repeating constitutional units comprises: a liquid chromatography device LC that has two columns arranged side by side and continuously injects a sample solution to carry out chromatography purification; an in-line that injects the sample solution continuously to the liquid chromatography device LC; a discharge line E of a purification recovery solution from the liquid chromatography device LC; a dilution solution reservoir B; a mixer M that connects the discharge line E to a dilution line from the dilution solution reservoir B to mix the solution; a solution reservoir R that receives the solution mixed and diluted at the mixer; and an out-line for connection to an in-line of a liquid chromatography device LC of the next stage from the solution reservoir R.
